Seeing too much predictability is problematic, but seeing too little can also be a problem, for example inappropriate "learned helplessness" can occur, whereby people feel disempowered because the world is seen as random. Recent findings demonstrated a bias in fundamental learning that may maintain these inappropriate beliefs about unpredictability. This bias is not anticipated by formal theories of learning. PurposeThis project aims to deepen knowledge of gastrointestinal physiology, and reveal the mechanisms by which the major gastrointestinal signalling molecule, serotonin, regulates gut peristalsis. Almost all of the serotonin in our body is made in the gastrointestinal tract where it controls many functions, including how our gut wall contracts during peristalsis. Proper control of gut peristalsis and the transit of material through our bowel is important for our health. This project expects to define how serotonin controls peristalsis, where in the bowel this serotonin comes from, how serotonin communicates with the nervous system in our gastrointestinal tract, and how the cells that synthesise gut serotonin respond to contraction to trigger the secretion of serotonin.

PurposeThis project aims to pinpoint the nature and timing of key steps in macropod history and to test how these link with major climatic and biotic changes. Macropods (kangaroos and relatives) are widely considered the marsupial equivalents to hoofed mammals on other continents, but we have a weaker understanding of how their evolution was shaped by environmental change. This project will combine palaeontology, anatomy and genetics to address questions such as how and why ancestral macropods descended from the trees and evolved bipedal hopping, and the upper size limits of the kangaroo -body plan-. This should improve our understanding of the long-term effects of climate change on marsupials, and provide a test of key placental-based evolutionary models.

PurposeThis project aims to identify the origins and movements of Australian archaeological ochre through the development of a novel tool combining genomic and chemical analysis. The geographic distribution of Australian ochre is closely linked to Aboriginal creation stories, while its physical distribution by people is evidence of cultural cooperation. Using this new archaeomicrobiological technique, the project aims to answer significant questions about past human behaviour, in terms of trade, cultural interactions, territoriality and colonisation. The method also has the potential to benefit traditional owners by contributing to repatriation projects. The collaborative detailed recording, sampling and analysis of ochre sources on traditional lands will also assist Aboriginal communities to manage this important aspect of their cultural heritage.

PurposeBowel cancer is a very common cancer in Australia. The best prevention strategy for people at increased risk of this disease is regular colonoscopies (every 3-5 years), however these are associated with risks, and places a large burden on limited healthcare resources. The majority of people do not need such intensive surveillance. Our proposal will test a novel way to reduce the frequency of colonoscopies needed through a personalised risk-assessment strategy using faecal occult blood tests.

PurposeThis project will use protein and nucleic acid biomarkers isolated from circulating exosomes (-ADMExosomes-) to characterise the activity of drug absorption, distribution, metabolism and elimination pathways that define drug exposure. In doing so, we will demonstrate the capacity of ADMExosomes to markedly enhance the ability to track and predict variability in drug exposure.

We present a new technology, called wireless optogenetics, whereby light, instead of drugs is used to stimulate the gastrointestinal tract without affecting any other organs. We present the first evidence that wireless optogenetics can stimulate the gut leading to increased transit of content in live mammals.

PurposeFaecal incontinence is embarrassing and can ruin a patient's quality of life. While the anal sphincters play an important role in controlling defaecation, patients can have normal sphincter function and still suffer severe incontinence. In this project we will use our unique recording techniques to identify an abnormal region of contractility in the distal colon of these patients and show that this contributes to the incontinence. These data will provide a new targeted region for treatment.

PurposeThe evolution of terrestrial animals from fish was one of the most significant events in our evolution, yet little is known about how the brain evolved during this transition. This project aims to investigate the major novelties acquired in the evolution of the early vertebrate brain in order to determine the functional reasons for such changes, as well as identifying the timing and environmental factors driving such changes. This project expects to generate new knowledge on the anatomy of the vertebrate brain with improved methods for reconstructing fossil brains to better understand our own neurological evolution. Expected outcomes include enhanced institutional collaborations within Australia, and between Australia, Canada and the USA.

PurposeParasites have been proposed to be drivers of population divergence, and ultimately speciation, yet the dynamics of this process are not well understood. This project will utilise new genomic techniques, novel hybrid zone analyses, and data on mate choice, to investigate the hypothesis that parasites drive population divergence through an interaction with immune response genes in the sleepy lizard Tiliqua rugosa. This species provides an unprecedented system, backed by 37 years of long term host-parasite and behavioural data, and recent genetic analyses. This project intends to produce significant data to allow an examination of the early stages of host-parasite evolution in action, providing novel insights into the speciation process.

PurposeThis project aims to generate unprecedented insights into the fangs of venomous snakes, focusing on elapids (taipans, tiger snakes etc). We will examine fang shape diversity, correlation with behavior and ecology, evolutionary history, and biomechanical properties. Data will be collected using cutting-edge micro-CT technology and analysed using 3D geometric morphometrics, computer simulations, and advanced phylogenetic techniques. This should greatly improve understanding of the evolution of venom fangs in all snakes. Other benefits include a large 3D reference database allowing identification of fossil fangs, with applications for studies of past climates, and a characterisation of fang biomechanics, relevant to biodesign and biomimicry.

The project will test whether and how utopian thinking stimulates a questioning of the status quo, moral engagement, and the formation of new groups to address social change: outcomes critical for a society to adapt and advance. Educators, government and non-government organisations can draw on project findings to re-engage a disaffected populace.

PurposeGlaucoma is a highly heritable complex disease and the leading cause of irreversible blindness worldwide. We used genetic techniques on a disease registry of individuals with glaucoma blindness to define risk factors associated with poor visual outcomes. The knowledge obtained from defining these risk factors will be used in a prospective longitudinal study to determine whether we can predict if individuals suspected of having glaucoma (glaucoma suspects) will progress to manifest vision loss.

PurposeThis project aims to deliver a proof-of-concept that allows visualisation of invisible DNA (latent DNA) into a quick, inexpensive and practical DNA collection method that will lead to DNA evidence being available in more cases. It will build upon a proof-of-concept method ready for transferal to forensic casework. This will allow DNA evidence recovery technicians to improve their hit rate in recovering latent DNA from real crime items, leading to more informative DNA profiles. Crime items that currently yield no genetic information will now be informative, assisting investigations of serious crimes or terrorist incidents.

The project is expected to generate fundamental knowledge using a unique combination of quantitative behaviour, neurophysiology, pharmacological intervention and biomimetic modelling. Expected outcomes include an increased understanding of neural mechanisms underlying sensory selectivity, the development of novel techniques, and enhanced capacity for interdisciplinary collaborations. PurposeChronic disease is the main reason Aboriginal people have poorer health outcomes compared to other Australians. The Antecedents of Renal Disease in Aboriginal Children (ARDAC) Study will generate critical new knowledge about when and how chronic kidney disease develops and progresses in Aboriginal people in the transition period from childhood to adulthood, and inform the development of sustainable, culturally appropriate interventions to reverse this trajectory.

PurposeThis study proposes a precision public health approach to eliminate STIs in far north Queensland. Our multi-disciplinary team will work generate evidence in the areas of population health, health services research and genomics, which will then be synthesized to inform precise targeted interventions to control STIs in the region. A community coalition will be established to oversee and lead this research process. We hope to be the first group globally to achieve elimination of STIs.

PurposeInterstitial cystitis is a severely debilitating bladder disorder with symptoms of urgency, frequency and pain. The mechanisms underlying these symptoms have not been determined but involve hyper-excitability of sensory nerves. This project will identify how hyper-excitability occurs in cystitis and the ion channels responsible. Our data will identify novel targets to treat visceral pain in patients with interstitial cystitis aimed at its root cause - sensory nerve endings of the bladder wall.

PurposeEndometriosis is a chronic painful disorder affecting ~10 percent of females worldwide, costing Australia $7.7 billion/year. The underlying mechanisms of chronic pelvic pain associated with endometriosis are poorly understood, with no efficacious treatment to the date. This project focuses in identify the molecular entities/mechanisms responsible for pain detection and transmission in endometriosis. This will provide novel therapeutics strategies and ultimately improve Endo patient's quality of life.
