PurposeThis project aims to synthesise the first examples of advanced biological imaging agents that can reversibly respond to the oxidative status of living cells. Novel mitochondrially-targeted, fluorescent probes will be derived from several well-established families of biological dyes through the introduction of a stable free radical within the parent structure. The design of the new imaging agents aims to both enhance retention in, and restrict the fluorescence response to, the mitochondria so that changes in oxidation and reduction can be monitored. The probes will provide an innovative new means to assess reactive species and associated oxidative stress, thus delivering a new methodology to aid research into mitochondrial chemical biology.

PurposeThis project aims to develop non-plasmonic, transition metal-based, photocatalysts with enhanced light absorption, to achieve irradiation controllable product selectivity in organic synthesis. The project demonstrates how hollow-particle morphology alloy nano'structures can enhance photocatalytic activity. Alloy structures such as iridium-nickel (IrNi), iridium-cobalt (IrCo) and iridium-tin (IrSn) nanoparticles with a hollow morphology, exhibit dramatically increased photocatalytic activity over their individual components, Ir, Ni, Co and Sn respectively. The project is expected to expand the application of photocatalysis and generate knowledge that can be used to design efficient photocatalysts from non-plasmonic metals. Intended benefits are the generation of new knowledge and capabilities in synthetic catalysis and applications in fields such as the conversion of solar energy to chemical energy.

PurposeThis project will enable automatic re-engineering of large enterprise applications to run in modern cloud environments as microservices. Microservices are the latest wave of service-based software, capable of exploiting the high performance and third-party integration opportunities made available through the cloud. The project will develop new techniques for analysing enterprise systems code and execution data, and making recommendations for restructuring suitable parts as microservices. These microservices manage individual business objects via sets of lightweight distributed computational operations. The outcomes will support progressive evolution of an enterprise system, into distributed microservices running in public clouds, while still being integrated with "backend" systems.

PurposeBy advancing pollutant transport modelling that use recent developments in drift sensors, this project aims to investigate water quality of estuaries and coastal rivers under pressure from urban growth. The project expects to generate a new capability for quantitative particle concentration predictions through detection and innovative analysis of Lagrangian Coherent Structures. The expected outcome is a new particle transport modelling framework and algorithms for shallow water systems, which effectively exploit extensive datasets becoming available from GPS-enabled drifters. This should provide significant benefits such as reliable assessment of possible effects of catchment and waterway changes on pollutant and sediment concentration which can impact waterway health.

PurposeThis project aims to improve measurement of retinal directionality, which is the reduction in brightness when a light beam's entry into the human eye is shifted from the centre to the edge of the pupil. This influences retinal image quality and can be used to measure changes in shape of the peripheral eye. This project will apply advanced technologies in holography, and heads-up displays to explore how retinal directionality changes during accommodation (focusing). The expected outcome is improved understanding of retinal stretching changes during focusing. The benefit is that the project will lead to advancements in retinal imaging.

PurposeThis project aims to investigate the structural diversity of lipids. Lipids are among the most structurally diverse of all the biomolecules and thus deciphering their many functions requires bio-analytical technologies capable of uniquely identifying and quantifying individual molecules in a milieu of many thousands of analogues. Mass spectrometry is the pre-eminent technique for contemporary lipid analysis but is challenged by the preference of certain lipids to ionise with a polarity that affords sensitive detection but does not permit detailed structure elucidation. This project will develop advanced instrumentation capable of on-demand polarity switching of ionised lipids such that the detection and interrogation of molecular structure can take place in the optimal charge state.

PurposeThis project aims to develop novel tests of visual function relevant to the modern night driving environment. Night driving is challenging for all drivers and has been linked to poor visibility under low light conditions. This project will characterise the visual challenges of the modern night driving environment, develop visual tests that incorporate the dynamic light levels typical of night-time roads and assess the association of these tests with night driving performance. The outcomes will contribute new knowledge regarding dynamic visual processing and the ageing visual system and will inform vision testing, potential interventions to improve visual function for night driving and reduce the dangers of night driving.

PurposeThis project aims to develop new mathematical tools for mapping tissue microstructural properties via the use of space-time fractional calculus methods.-In magnetic resonance imaging, mathematical models and their parameters play a key role in associating information between images and biology, with the overall aim of producing spatially resolved maps of tissue property variations. However, models which can inform on changes in microscale tissue properties are lacking. The tools developed by this project will be used to generate new magnetic resonance image based maps to convey information on tissue microstructure changes in the human brain. Additionally, the mathematical tools developed will be transferable to other applications where diffusion and transport in heterogeneous porous media play a role.

PurposeThis project aims to better understand how young adults use, communicate about and experience mobile phone sports betting applications. Gambling generates significant health and social harms in Australia. Yet there is little research on the use of betting apps, even though sports betting is the fastest growing segment of the gambling market. This project intends to examine how use of sports betting apps is becoming established as everyday social practice normalising problem gambling. The findings will enhance understanding of the social contexts of sports betting, and inform gambling policy and programs leading to better health and social outcomes.

PurposeThis project aims to quantify the impacts of native and non-native animal consumer removal by examining food webs in endangered grasslands. Grasslands are Australia's largest biome yet land-use changes, livestock and invasive plants, have altered entire food webs, including the integrity of ecosystem services such as nutrient cycling. Measuring disrupted food webs in field experiments, then modelling the impacts of mammals to invertebrates, will assist managers in making more effective decisions relating to ecosystem integrity. Understanding the consequences of biodiversity loss including implications for ecosystem resilience is crucial to Australia's future food production, carbon sequestration and hydrological flows.

PurposeThis project aims to examine the interaction of fundamental two-dimensional patterns such as spots and stripes in reaction-diffusion equations, by developing and extending mathematical techniques. These fundamental planar structures form the backbone of more complex patterns and are, for example, observed in models that describe the propagation of impulses in nerve axons and the formation of vegetation patterns. The future impact of this research will have economic and environmental benefits. For example, the project will develop a deeper understanding of interacting patterns that will provide insights into the role of vegetation in ecosystems that are undergoing desertification.

PurposeThis project aims to produce new knowledge to foster inclusion of people with disabilities in local community planning practice in regional areas. Exclusion of people with disabilities in local communities persists despite the national disability reform agenda. This project will place spatial justice thinking and critical disability theory within a community planning and development context to examine the concept as a scaled phenomenon. Using participatory research methods, the project will directly engage persons with disabilities and community planners in co-producing an adaptive model of practice. The project outcomes will help to create more inclusive, healthy, and resilient communities that enable people with disabilities to lead ordinary and fulfilling lives.

PurposeThis project aims to investigate the performance and wellbeing of introverted leaders. It intends to test a theoretical model of leader performance and wellbeing which recognises that introverts regularly need to act out of character, that is, extraverted, in order to perform competently in leadership positions. The project proposes that the necessity for introverted leaders to act extraverted will compromise their effectiveness and make them vulnerable to low wellbeing. Expected outcomes from this project include a better understanding of the performance and wellbeing of introverted leaders. Intended benefits for introverted leaders include demonstrated efficacy of affective forecasting intervention strategies.

PurposeThis project aims to quantify the interdependencies of the psychosocial risk factors the construction workforce is exposed to and contribute to developing strategies for improving the mental health and safety of the Australian construction industry. The suicide rate of Australian construction workers is 84 per cent higher than non-construction workers due to the high psychosocial risks involved. The quantification of these psychosocial risk factors and their interdependencies will enable effective strategies to be designed. The long-term impacts of this project include improving the mental health of the construction workforce, reducing suicide rates and mental health work compensation, enhancing employee productivity, and reducing accidents and injuries. This project will make Australia a global leader in mental health and safety of the construction industry.

PurposeCADASIL is a cerebral small vessel disease which is one of the most common heritable cause of stroke and vascular dementia in adults. Genetic diagnostic testing for NOTCH3 mutations causative of CADASIL only identifies mutations in ~20 percent of patients which suggests that additional genes and mutations are responsible for CADASIL. By utilising whole exome sequencing on CADASIL patients which have no causative NOTCH3 mutations, I aim to identify new genes that are causative of CADASIL.

PurposeIn order to improve the treatment of children's burn injuries, we need to better diagnose burn wound severity and predict which patients are at risk of developing poor long-term outcomes. By examining the proteins and immune cells in burn patient samples, we can identify markers that are related to poor healing outcomes. This will help us to develop better clinical prognostic tools for burn wound assessment and better patient outcomes.

PurposeMultiple myeloma is a type of blood cancer. Despite recent advances in treatment, multiple myeloma remains incurable. We demonstrated that the myeloma-associated inflammation critically drives disease progression by generating an environment that limits anti-myeloma immune responses. PurposeGenome-wide association studies for psychiatric disorders have been highly successful in identifying risk loci. While the effects of these loci are assumed to be within the brain, it is unclear which brain regions should be studied. We propose a series of analyses to help clarify which loci influence the brain and where these effects are located. We will use the results of these analyses to develop a searchable database mapping the effects of brain expressed loci on key brain regions.

PurposeRespiratory syncytial virus bronchiolitis kills up to 200,000 infants every year, and increases the risk of asthma. In an Argentinian cohort it was found that poor maternal diet predisposes to severe bronchiolitis. Replicating this in mice, we have linked this finding to alterations in the gut microbiome and key immune cell populations. Here, we will use clinical samples and our mouse model to identify new microbial-derived molecules that drive immune development and protect against disease.

PurposeIn Australia around 25 percent of clinically-confirmed pregnancies end in miscarriage. While up to 60 percent are likely due to chromosomal or structural anomalies in the developing embryo, the causes of the remainder are typically unknown. PurposeAcute myeloid leukaemia (AML) is a very aggressive blood cancer that kills about 1,000 Australians every year. AML patients are treated with chemotherapy, however, most patients are not cured by chemotherapy alone and the disease eventually comes back (relapses). We are testing a new type of treatment (imetelstat) for patients with AML that targets the genetic material (DNA) within the leukaemia cells. Our work has shown that this new type of treatment may prevent relapse after chemotherapy.

PurposeBowel cancer is a very common disease that will be diagnosed in 1/12 Australians living to age 85 and leads to over 4000 deaths annually. In this application, we are proposing to research a specific type of bowel cancer that has been associated with a particularly poor outcome. We will use a combination of pathological, genomic and cell biology techniques to better understand these cancers and discover new ways to treat them.

PurposeBone marrow transplantation can cure leukaemia but is often complicated by graft-versus-host disease (GVHD), which greatly reduces quality- of-life. A type of immune cell, called regulatory T cells (Tregs), may help. We will expand donor Tregs in the laboratory and infuse them into patients with chronic GVHD. We will gene-mark the cells so that we can understand what happens after infusion. We will also investigate whether interleukin-2 given at the same time can make the Tregs more effective.
ActivityGNT1163111 | A phase I clinical study of adoptive transfer of regulatory T cells (Tregs) and low-dose interleukin-2 (IL-2) for the treatment of chronic graft-versus-host disease (GVHD): gene-marking to inform rational combination therapy
