PurposeAnxiety is a debilitating symptom affecting many people with autism. However, we do not know how effective and how well tolerated are the medications, which are used to treat anxiety in the general population, when prescribed to people with autism. To answer this question we propose a clinical trial of one anxiety medication (sertraline) across five sites in two countries. We will recruit over 300 adults with autism and half with receive the active drug and half a placebo or dummy medication.

For thousands of years we have used microbes to create bread, wine, cheese. Now, our Centre will pioneer new approaches to the design of synthetic microbes, enabling the development of custom-designed microbial communities, synthetic organelles and new to nature biological pathways and enzymes. CoESB will combine engineering with molecular biology to design and construct novel biological systems that can convert biomass from agriculture or waste streams to biofuel, bioplastics and other high-value chemicals.

PurposeThe success of modern lung cancer radiotherapy relies on four-dimensional computed tomography (4DCT) scans to account for breathing-induced tumour motion. Currently, 90% of 4DCT scans suffer severe image errors that limit the treatment accuracy and reduce the chance of tumour control by up to one third. This project will translate high accuracy, real-time adaptive 4DCT from a software prototype to clinical reality, enabling more accurate treatments and better outcomes for up to 5,000 Australians/year.

PurposeThere are no effective treatments for patients with relapsed glioma. In the subset of patients with isocitrate dehydrogenase (IDH)-mutant glioma, there is promising biologic rationale for, and potential synergy, combining rucaparib (which inhibits DNA repair) and nivolumab (which augments the anti-cancer immune response). This combination has been previously reported to be safe. This study will test the effectiveness of rucaparib and nivolumab against relapsed IDH-mutant glioma.

PurposeMerkel cell carcinoma (MCC) is a rare but highly aggressive type of skin cancer that has a much worse survival than melanoma. MCC mainly affects the elderly and those with weak immune systems and extensive sun exposure. Most patients present with early stage MCC, usually treated with surgery +/- radiotherapy (RT). This study will investigate whether the use of immunotherapy, shown to be effective in advanced MCC, can prevent the cancer returning after surgery, when given with RT.

PurposeSurvival for children with relapsed and recurrent brain tumours is poor, and there have been few new treatment options. However, recent research shows that these tumours have a general change in their DNA (methylation), and altering methylation may stop the tumours. PurposeSJELiOT is a phase 1 clinical trial that explores the combination of a new drug called LY2606368 with other chemotherapy drugs used to treat the childhood brain cancer medulloblastoma. PurposeNecrotising enterocolitis is a rare gut disease that occurs in 1-5/10,000 newborns. A third of infants will die and many survivors will suffer long-term health and neurodevelopmental impairment. A link between blood transfusions for anaemia and the incidence of NEC has been reported. This world-first neonatal point-of-care trial, innovatively embedded in a national clinical quality registry, will test if withholding feeds for 12 hours around transfusion time will prevent this serious disease.

PurposeThere is an unmet need to reduce the mortality and disablity of the most severely injured trauma patients. Erythropoietin (EPO), a hormone which controls the production of red blood cells, also plays a key role in regulating our bodies response to injury and trauma. Previous work strongly suggests that EPO may decrease death and disability of critically ill trauma patients. We will perform the highest quality clinical trial to determine if EPO improves the lives of those critically injured.

PurposeThe phase 3 randomised controlled trial aims to improve the outcome of Follicular Lymphoma patients who remain PET positive after initial immunochemotherapy (who have demonstrated to have a poor outcome), and minimise further treatment for good prognosis patients who become PET negative. However, its utilisation has been decreasing due to high infection burden. The TEACH-PD study will determine whether delivery of a standardised training curriculum designed for PD nurse trainers and patients reduces patients- PD-related infection risk and improves their capacity to participate in usual activities.

Our U.K. and European colleagues have developed a new trial, the FaR-RMS study, which includes optimising current treatments, as well as testing the latest promising agents. This study is designed to answer a number of questions within the one trial, and new drugs can be rapidly added. Opening this trial in Australia gives new hope to all patients battling RMS.

PurposeMantle Cell Lymphoma (MCL) has short survival and new treatments are urgently required. Our group pioneered the ibrutinib-venetoclax combination in MCL in the AIM1 study, reporting a 71 percent complete remission rate. In this proposal, we will use the knowledge from AIM to identify resistant patients apriori, and introduce the new drug navitoclax. For all patients, we will include the monoclonal anti-CD20 antibody. These results will be compared against the results of the AIM1 study.

PurposeThe ACHIEVE Study aims to improve the poor outcomes of patients with kidney failure, which are mostly driven by heart disease. It will study an existing drug, spironolactone, that is proven to benefit patients with heart failure. Spironolactone is rarely used in kidney failure as few studies have tested it, and there are concerns about its side effects in this patient group. This large, international study will provide reliable evidence as to whether this promising treatment works.
